Transfer approximately 200 μl of the sample mixture into a clean test tube for testing. Insert the test strip vertically into the test tube containing the sample mixture. The light red protein A-colloidal gold marker on the test strip will dissolve, and the IgG in the sample (the reaction principle of other antibodies in the sample, such as IgA and IgM, is the same as that of IgG) will combine with the protein A/colloidal gold particles to form an "IgG-protein A-gold" complex. The "IgG-protein A-gold" complex will move upward along the test strip and first reach the detection area coated with HIV antigen. If the sample contains HIV antibodies, the "IgG-Protein A-Gold" complex will bind to the antigen and remain on the test line, forming a light red line, which indicates a positive result. The depth of the line color is not proportional to the amount of antibodies in the sample. If there is no colored line in the test area, it means that the sample does not contain HIV antibodies, and the free "IgG-protein A-gold" complex continues to move to the top of the test paper and reaches the control area of the test paper. The control area was coated with goat anti-human IgG F(ab')2 fragment and fixed on the control line of the test strip. The "IgG-Protein A-Gold" complex will bind to the goat anti-human IgG F(ab')2 fragment and be enriched on the control line, forming a light red line. The appearance of the control line proves that the test strip is functioning normally. Regardless of whether the sample contains HIV-1/2 antibodies, in a valid test, a light red to purple control line should appear in the control area of the test strip. The sample continues to move through the control zone and into the absorption zone. The function of the absorption zone is to adsorb the remaining "IgG-protein A-gold" complex, make it move within the test strip, and eliminate the background color. The result can be read 20 minutes after the test strip is inserted into the diluted sample mixture, but no later than 45 minutes.
